Domperidone belongs to a class of drugs called the anti emetics. It is a dopamine D2 and D3 receptor antagonist. It speeds gastrointestinal peristalsis
Adults and adolescents (12 years of age and older and weighing 35 kg or more): One 10mg tablet up to three times per day with maximum dose of 30 mg per day.
- Domperidone's elimination half-life is prolonged in patients with significant renal impairment. Dosing frequency of domperidone should be lowered to once or twice daily for recurrent administration, depending on the degree of the impairment. It's also possible that the dose will need to be lowered.
- The drug domperidone has been linked to an increase in the QT interval on an ECG. There have been extremely few reports of QT prolongation and torsades de pointes in patients on domperidone during post-marketing surveillance. Patients with confounding risk factors, electrolyte imbalances, and concurrent medication were included in these publications, all of which could have been contributory factors.
- Domperidone was linked to an elevated incidence of severe ventricular arrhythmias or sudden cardiac death in epidemiological investigations.
Contraindication
- Known hypersensitivity to any component of the medication.
- Co-administration with QT-prolonging drugs, at the exception of apomorphine, such as-

Domperidone is contraindicated in the following situations:
- In patients with moderate or severe hepatic impairment
- In patients who have known existing prolongation of cardiac conduction intervals, particularly QTc, patients with significant electrolyte disturbances or underlying cardiac diseases such as congestive heart failure
- Prolactinoma
- When stimulation of gastric motility could be harmful e.g in patients gastro-intestinal hemorrhage, mechanical obstruction or perforation.
