Tazobactam

Tazobactam is classified as a beta-lactamase inhibitor. It acts as a mechanism-based inhibitor of beta-lactamases, enzymes produced by certain bacteria that confer resistance to beta-lactam antibiotics. Tazobactam, when combined with beta-lactam antibiotics, enhances their efficacy by preventing the degradation of the antibiotic by beta-lactamases.

Tazobactam is indicated for use in combination with certain beta-lactam antibiotics to treat infections caused by susceptible bacteria. It is often used in combination with piperacillin, a broad-spectrum penicillin, to broaden the spectrum of antibacterial activity.

The recommended dose typically involves the co-administration of tazobactam with a beta-lactam antibiotic. Dosage and duration depend on the severity of the infection and the specific antibiotic used.

Adverse reactions associated with tazobactam are-

  • Nausea
  • Rash
  • Headache
  • Vomiting
  • Increased liver enzymes
  • Injection site reactions
  • Hypersensitivity reactions (e.g., anaphylaxis)
  • Hypersensitivity Reactions: Monitor patients for signs of hypersensitivity reactions, including rash, fever, and anaphylaxis. Discontinue treatment if an allergic reaction occurs and initiate appropriate medical therapy.
  • Clostridium difficile-associated Diarrhea (CDAD): Consider CDAD in patients presenting with diarrhea after antibiotic use. Discontinue Tazobactam if CDAD is suspected or confirmed and initiate appropriate treatment.
  • Renal Impairment: Adjust dosage in patients with renal impairment to prevent accumulation of the drug and potential toxicity. Monitor renal function regularly.
  • Seizures: Use with caution in patients with a history of seizures or central nervous system disorders. Seizures have been reported in patients treated with beta-lactam antibiotics, including tazobactam.
  • Superinfection: Prolonged use may result in overgrowth of nonsusceptible organisms, including fungi. Monitor patients for superinfections and institute appropriate therapy if necessary.

Contraindication

Tazobactam is contraindicated in patients with a known hypersensitivity to tazobactam, beta-lactam antibiotics, or any component of the formulation.
