Solifenacin Succinate

Solifenacin Succinate is a muscarinic antagonist.

Solifenacin Succinate indicated for the treatment of overactive bladder with symptoms of urge urinary incontinence, urgency and frequency. 

5 mg tablet taken once daily, and if well tolerated may be increased to 10 mg once daily.

Do not exceed 5 mg tablet once daily in patients with:

  • severe renal impairment [Creatinine Clearance] (CLcr <30 ml/min)
  • moderate hepatic impairment (Child-Pugh B)
  • concomitant use of potent CYP3A4 inhibitors 

 

The following adverse reactions are reported with the use of Solifenacin Succinate-

  • Dry mouth
  • Constipation
  • Dyspepsia
  • Abdominal pain
  • Nausea
  • Diarrhea
  • Urinary tract infection
  • Dizziness
  • Asthenia
  • Dry eyes
  • Because of the risk of urinary retention, Solifenacin extended-release tablets should be used with caution in patients with clinically significant bladder outflow obstruction.
  • Because of the risk of gastric retention, Solifenacin should be used with caution in patients with gastrointestinal obstructive disorders. Solifenacin, like other anticholinergic drugs, can reduce gastrointestinal motility and should be used with caution in patients with severe constipation, ulcerative colitis, or myasthenia gravis.
  • Solifenacin should be used with caution and only when the potential benefits outweigh the risks in patients with narrow-angle glaucoma.
  • Use of Solifenacin is not recommended in patients with severe hepatic impairment.
  • Angioedema and anaphylactic reactions: Reports of angioedema of the face, lips and/or larynx, in some cases occurring after the first dose, have been described. Anaphylactic reactions have been reported rarely.
  • QT Prolongation: Use with caution in patients with a known history of QT prolongation or patients who are taking medications known to prolong the QT interval.
  • Central Nervous System Effects: Somnolence has been reported with Solifenacin. Advise patients not to drive or operate heavy machinery until they know how Solifenacin affects them

Contraindication

Contraindicated in patients who have demonstrated hypersensitivity to the Solifenacin.

Solifenacin is contraindicated in-

  • Urinary retention
  • Gastric retention 
  • Uncontrolled narrow-angle glaucoma
